重组人肿瘤坏死因子-α(TNF-α)(Human TNF-α protein)是一种通过大肠杆菌表达系统制备的高纯度细胞因子,采用先进工艺纯化,适用于细胞培养研究中的体外细胞毒性试验等应用。
1975年Carswell等人首次发现肿瘤坏死因子在介导肿瘤出血性坏死中的关键作用,其中TNF-α占TNF总生物活性的70-95%。该因子主要由活化巨噬细胞和T淋巴细胞分泌,通过结合特定受体触发细胞凋亡通路。本产品采用人源TNF-α可溶形式(Val77-Leu233)的161个氨基酸序列,经大肠杆菌重组表达制备。其生物活性经标准化验证:在放线菌素D存在条件下,通过L929小鼠纤维肉瘤细胞毒性试验测定,显示25-100 pg/ml的ED50值,确保其在细胞实验中的功能可靠性。
